Preparation and Study of Solid Dispersions of Meloxicam

Meloxicam is a non-steroidal anti-inflammatory drug that is practically insoluble in water. The aim of the present work was to study the influence of the preparation method of solid dispersions and the nature of the carrier on the solubility of meloxicam in vitro. Solid dispersions of meloxicam using water-soluble polymers were prepared by solvent-removal and co-melting methods. The practical yield of the obtained solid dispersions was evaluated to confirm the suitability and efficiency of the manufacturing technique. The microcrystalline pattern was shown. A comparative characterization of meloxicam solid dispersions was given. The solubility of the obtained solid dispersions was studied in comparison with the native substance and physical mixtures with polymers by UV-spectrophotometric analysis of the meloxicam concentration in the dissolution medium (phosphate buffer, pH 6.86). The possibility of increasing the water solubility of meloxicam by 5.5 times when using polyvinylpyrrolidone as a carrier polymer was shown. Solid dispersions in the form of milled powder showed improved technological properties as compared to the native substance. The proposed methods of obtaining solid dispersions will optimize the production process of drugs based on them and produce drugs with increased solubility of the active ingredient in biological fluids, which in turn will help to increase the bioavailability of meloxicam in the therapy of inflammatory diseases of the musculoskeletal system.
