<p><b>Objective.</b> To study the associations of single-nucleotide polymorphisms of the <i>NOS1AP</i> gene with the duration of the QT interval in patients with schizophrenia taking antipsychotics. <b>Materials and methods.</b> A total of 168 patients (78 men and 90 women) diagnosed with schizophrenia were examined. The study patients received basic antipsychotic therapy. Standard 12-lead electrocardiograms were recorded on admission to hospital. The QTc interval was calculated using the Bazett formula. Three single nucleotide polymorphisms of the <i>NOS1AP</i> gene (rs12029454, rs10494366, and rs12143842) were selected for genotyping. <b>Results.</b> Application of the Spearman test identified a statistically significant correlation between chlorpromazine equivalent (CPZeq) and the duration of the QTc interval in the group of men (r<sub>s</sub> = 0.262; <i>p</i> = 0.021; <i>p</i><sub>bonf</sub> = 0.042). In addition, correlation analysis showed that in men with the AA and AG rs12029454 (r<sub>s</sub> = 0.717; <i>p</i> = 0.00025; <i>p</i><sub>bonf</sub> = 0.0015), GG and GT rs10494366 (r<sub>s</sub> = 0.381; <i>p</i> = 0.008; <i>p</i><sub>bonf</sub> = 0.048), and TT and CT rs12143842 (r<sub>s</sub> = 0.389; <i>p</i> = 0.003; <i>p</i><sub>bonf</sub> = 0.018) genotypes, CPZeq correlated positively with QTc interval duration. No significant correlations were found in the group of women. <b>Conclusions.</b> Polymorphic variants rs12029454, rs10494366 and rs12143842 of the <i>NOS1AP</i> gene are associated with QTc interval duration in male patients with schizophrenia in response to antipsychotics. Carriership of the AA/AG rs12029454, GG/GT rs10494366, and TT/CT rs12143842 genotypes in men contributes to prolongation of the QTc interval and the development of adverse cardiovascular events during antipsychotic therapy.</p>
