Background <p>Postpartum anestrus (PPA) is a significant cause of infertility in dairy cattle, often delaying the resumption of ovarian cyclicity. Anti-Müllerian hormone (AMH) serves as a biomarker of endocrine activity and ovarian follicular reserve. This study investigated the association between AMH gene polymorphisms and prolonged PPA in Holstein-Friesian cattle. <i>Methods and Results</i> A total of 305 cows were categorized into three groups based on reproductive history of anestrum and rectal ultrasonography: cyclic, PPA 6–12 months, and PPA more than 12 months. Blood samples were collected in vacutainers coated with EDTA- and kept at -20&#xa0;°C for subsequent genomic DNA isolation. Data were recorded for age at first calving (AFC), calving interval (CI), calf birth weight (CBW), milk yield (MY), and days in milk (DM) for all animals. Genotype and allele frequencies for Exon 1 and Exon 5 of the AMH gene were analyzed using PCR-SSCP. PCR amplification revealed bands of 343&#xa0;bp for Exon 1 and 253&#xa0;bp for Exon 5. Five distinct SSCP patterns were identified for Exon 1, while Exon 5 exhibited only two patterns. Sequence analysis confirmed five genotypes (GG, CC, AA, CA, and GT) in Exon 1, with three single-nucleotide polymorphisms (SNPs) identified: a synonymous mutation, rs462782396 (Chr7.21401756G &gt; T), and two previously unreported SNPs, Chr7.21401691&#xa0;C &gt; A and Chr7.21401708&#xa0;C &gt; A. Exon 5 exhibited two genotypes (TT and CT) and harbored a missense mutation, rs43505518 (Chr7.21403549T &gt; C). <i>Conclusions</i> Significant associations were observed between AMH gene polymorphisms and prolonged postpartum anestrus (PPA) in cattle, particularly with respect to AFC and DM.</p>
