Background <p>During the last decade, commercial companies have increased the number of Short tandem repeat (STR) loci in their Human identification (HID) kits to improve the forensic conclusions. For instance, Promega Corp. offers the PowerPlex Fusion 6&#xa0;C System and VersaPlex 27PY systems (Promega Corp.) that analyze the same 22 STRs and three Y-chromosome STRs (Y-STRs), plus the SE33 and D6S1043 <i>loci</i>, respectively. Thus, more population studies are needed for paternity testing and criminal interpretation, principally.</p> Methods and results <p>We genotyped 559 and 606 unrelated Mexican volunteers from two nearby western states with the PowerPlex Fusion 6&#xa0;C and Versaplex 27PY Systems, respectively. We computed allele frequencies and forensic parameters, besides to assessing the Hardy-Weinberg equilibrium (HWE) and linkage disequilibrium (LD) agreement for the total western population sample (<i>n</i> = 1165). Interpopulation analysis, including American ethnic groups, supported the combination of these western population samples for the STRs shared between HID kits. The combined PD and PE for the 23 STRs were virtually 100% and 99.999999993%, respectively. The three Y-STRs (DYS391, DYS576, and DYS570) exhibited 137 different haplotypes among 992 males, resulting in a haplotype diversity of 97.7%. The genetic relationships and population structure among Mexican populations supported a clinal differentiation between two main geographic areas: (1) North and West; and (2) Center, South, and Southeast.</p> Conclusions <p>This study provides a comprehensive Mexican STR database for two understudied HID kits, which are helpful for forensic casework interpretation in this country.</p>
