<p>N,N,N’, N’-Tetraoctyldiglycolamide (TODGA) is a pivotal extractant for minor actinide separation in nuclear fuel cycles. This work presents a novel one-step synthesis method of TODGA using 2-(7-Azabenzotriazol-1-yl)-N,N,N’,N’-tetramethyluronium hexafluorophosphate (HATU) as the coupling agent. The target product was obtained under ambient temperature and pressure within 1.5 h, with a yield of &gt; 94%. Its purification required only simple extraction, affording a purity of &gt; 95%. Scaled-up synthesis enabled kilogram-scale production of TODGA. Extraction tests confirmed that the synthesized TODGA exhibits excellent efficacy in the sequestration and separation of Am(III) and Eu(III), thus verifying its promising application prospects in nuclear waste treatment.</p>
