<p>This study explores the impact of build orientation on the mechanical, surface, and bioactivity properties of additively manufactured PLA and Re-PLA filaments. PLA and Re-PLA were printed in six orientations at three different filling ratios, with a filament diameter of 1.75&#xa0;mm, layer thickness of 0.2&#xa0;mm, and a grid filling shape. The materials were characterized through tensile, flexural, surface roughness, SEM analysis, degradation, and bioactivity (antibacterial and biocompatibility) tests. Results show that build orientation significantly affects the mechanical properties of PLA, with ZXY-ZYX binary groups exhibiting higher bending strength than other orientations. Re-PLA was found to break more uniformly than PLA. No bacterial activity was observed against <i>S. aureus</i> or <i>E. coli</i> for both materials. Cell viability decreased across all filling ratios in Re-PLA, with a general reduction in cell viability after four days of culture compared to day one. No degradation was detected in PLA and Re-PLA filaments at 20%, 40%, and 60% filling ratios. This study aids in selecting optimal factors like structure orientation and filling ratio for various applications and emphasizes the use of recycled biopolymers from renewable resources, such as PLA, over petroleum-based biodegradable polymers.</p>
