<p>Floridoside are&#xa0;galactosylglycerol&#xa0;(GG) found in red algae and are known for their diverse physiological roles and bioactivities. In this study, we characterized isofloridoside in dried sheets of nori <i>Pyropia yezoensis</i> which was cultivated in the Ariake Sea, Japan. Isofloridoside was extracted and purified from color-faded grade nori, crystallized and its structure confirmed by GLC, <sup>13</sup>C-NMR, and ESI-TOF mass spectrometry. Quantitative GLC analyses of floridoside, D-isofloridoside, and L-isofloridoside were carried out using eight different grades of dried nori sheets, the thalli of which were cultivated in the same area of the Ariake Sea during the same cultivation period. Isofloridoside was shown to be the dominant constituent (82–88% of total GG), with the L-isomer accounting for approximately 70% of the isofloridoside. The GG content varied significantly among the different grades. Lower-rated nori, such as the color-faded grades, had a higher GG content (over 120&#xa0;mg&#xa0;g<sup>−1</sup> dried nori), whereas higher-rated nori, such as grades 1 and 2, had a lower content (approximately 30&#xa0;mg&#xa0;g<sup>−1</sup> dried nori).</p>
