Modulation of inflammatory and oxidative stress pathways by polyphenol-rich Pleurospermum candollei extract in carrageenan-induced paw edema in rats

Pleurospermum candollei is a locally used vegetable traditionally known for its medicinal properties in alleviating pain and joint discomfort. The present research was designed to assess the anti-inflammatory potential of P. candollei butanol extract (PCB) in the carrageenan-induced paw edema rat model.
MethodsHPLC–DAD technique was employed for phenolic detection in P. candollei. In vitro antioxidants and anti-inflammatory activities were examined using various assays. In vivo anti-inflammatory potency of PCB was examined against carrageenan-mediated paw edema, while indomethacin was used as a positive control. Medicinal qualities of PCB were further analyzed through assessment of antioxidant enzymes, oxidative radicals, and myeloperoxidase (MPO) activity in paw tissues. qRT-PCR analysis investigated the gene expression of various inflammatory markers to confirm anti-inflammatory potential of PCB.
ResultsPCB exhibited high concentrations of phenolics and showed in vitro antioxidant activities against free radicals. PCB decreased the concentration of free radicals and malondialdehyde in paw tissues while increasing in vivo antioxidant levels and MPO activity. Furthermore, PCB administration downregulated mRNA expression of inflammatory genes (IL-6, TNF-α, and IL-1β) and cyclooxygenase-2 (COX-2) compared to control.
ConclusionThese findings highlight the strong anti-inflammatory potential of P. candollei by enhancing enzymatic antioxidant activities and reducing inflammatory marker levels. The results further support the therapeutic value of natural plant-based compounds in the development of novel anti-inflammatory drugs.
