Deciphering genetic diversity and population structure in Crossandra (Crossandra infundibuliformis L.) genotypes based on SSR markers

Crossandra infundibuliformis (L.) is a flowering perennial shrub with extensive application in ornamental uses, such as pot plants, flower beds, loose flowers and pharmaceuticals, making deciphering its genetic diversity and population structure essential for identifying superior and distinct germplasm. The present study carried out to analyze the genetic diversity of Crossandra infundibuliformis using simple sequence repeat (SSR) markers. In this study, Crossandra genotypes were screened for amplification of 59 cross-species simple sequence repeat (SSR) markers. Twenty-nine polymorphic markers were utilized to estimate the genetic diversity in 20 Crossandra genotypes. A total of 102 polymorphic bands were produced by 29 primers, with an average of 3.52 bands per primer. The polymorphic information content (PIC) varied between 0.008 and 0.994. Neighbor joining (NJ) based cluster analysis revealed a dendrogram with clusters that coincided mostly for phenotypic traits, primarily flower colour, form, plant stature, peduncle color, and inflorescence color. The population structure analysis also revealed that the population was divided into two genetic groups, as shown by the maximum ΔK of 2, which was correlated with neighbor joining (NJ) clustering. This study emphasizes the novelty of Crossandra infundibuliformis as it was the first to report on its molecular diversity and population structure. These findings strengthen efforts to conserve gene pool, aid in the identification of distinct genetic lines, and facilitate their application in programs for varietal improvement.
