<p>Olive oil production is a key aspect of assessing the genetic structure of olive cultivars in Mansehra, since olive oil is a valuable commodity with growing demand, selecting superior cultivars can enhance local farmers. Six different genotypes of olive (<i>Olea europaea</i> L.) were obtained from the National Tea and High-Value Crops Research Institute in Mansehra, from Shinkiari and Hathi Mera in Khyber Pakhtunkhwa, Pakistan, to assess their genetic diversity. Genomic DNA was extracted from fresh leaves using a modified CTAB method. For RAPD-PCR analysis, 39 random primers were tested, of which 32 successfully generated highly polymorphic and distinct bands, averaging 3.3 polymorphic bands per primer. The fragment sizes ranged between 500 and 1000&#xa0;bp. Among the tested primers, OPA-18 produced the highest number of bands (16), while OPC-05 generated the lowest (9) bands. The highest genetic distance (69.31%) was recorded between the Picual and Corotina cultivars, whereas the lowest genetic variation (15.63%) was observed between the Nebali and Megaran cultivars. The application of RAPD-PCR markers provided valuable insights into the genetic relationships among different olive cultivars, emphasizing the considerable genetic divergence between Picual and Corotina and the relatively close genetic similarity between Nebali and Megaran. The genetic structure of olive cultivars in Mansehra, Pakistan, provides crucial insights into the diversity and adaptability of these crops. This knowledge enables the identification of superior cultivars for improved yield and disease resistance. Ultimately, such genetic assessments contribute to enhancing olive production and sustainability in the region.</p>
