<p>The date palm (<i>Phoenix dactylifera</i> L.) is a prominent fruit tree belonging to the genus Phoenix and the Arecaceae family. Pakistan, home to over 300 cultivars, exhibits remarkable biodiversity particularly in Khairpur region. Rich in nutrients and antioxidants, dates offers significant health benefits largely attributed to their phenolic compounds and antioxidant content. The selection of pollen parents plays a crucial role in dtermining fruit quality and yield, a phenomenon known as metaxenia. This study focused on khudrawi cultivar to assess the impact of nine different male pollen sources on the biochemical traits. The results revealed that these pollen sources exert diverse effects on biochemical parameters, including soluble sugar content, 2,2-diphenyl-l-picrylhydrazyl content, and phenolic content. Enzymatic activities such as catalase, superoxide dismutase, and protease also varied significantly among different pollen sources, further highlighting their influence on fruit quality and biochemical prperties. Among the tested pollen sources, M8 (Male8) demostrated most favorable impact, followed by M7 (Male7), M3 (Male3), M5 (Male5), and M4 (Male4). A stronge positive correlation was observed among various biochemical traits, underscoring their importance in metaxenia research and breeding programs aimed at improving both fruit quality and yield. Furthermore, genetic analysis using simple sequence repeats markers identified 66 loci with variable major allele frequencies, particularly in mpdCIR015 and DP159 primers. These findings emphasize the necessity of careful pollen parent selection and genetic analysis to enhance date palm cultivation and preserve its cultural, agricultural, and economic importance.</p>
