A holistic approach to pharmacological profiling and phytochemical analysis of the endemic Scutellaria orientalis L. subsp. bicolor (Hochst.) J.R.Edm (Lamiaceae)

This study aimed to evaluate the chemical compositions of the extracts of varying polarities of the endemic Scutellaria orientalis L. subsp. bicolor (SOB) and assess their biological activities. The relationships between various biological activities were visualized using multivariate methods such as principal component analysis (PCA), hierarchical cluster analysis (HCA), and triangular heatmaps with Pearson correlation matrix analyses. The highest extraction efficiency was observed with the aquoeus extract (20.31%). The investigations of the phenolic components of different fractions of SOB were optimized by identifying and quantifying of 36 phytochemical fingerprint compounds using LC–ESI–MS/MS analysis. Shikimic acid, scutellarin and baicalin were identified as the main components in varying amounts. Total phenolic (TPC) and flavonoid (TFC) contents were the highest in the aqueous SOB extract (25.78 ± 2.21 μg PEs/mg extract and 43.45 ± 0.31 μg QEs/mg extract, respectively). The antioxidant activity of the extracts, evaluated using DPPH, ABTS, and CUPRAC, displayed superior results for the aqueous SOB and SOB-acetone extracts. Remarkable values were observed for DPPH (IC50 value: 57.09 ± 0.29 μg/mL, SOB-EtOH extract), ABTS (IC50 value: 26.22 ± 0.26 μg/mL, SOB aqueous extract) and CUPRAC (A0.5 value: 83.43 ± 0.69, SOB-EtOH extract). Despite similar trends in enzyme-inhibitory activity, SOB-hexane and SOB-aqueous extracts showed the highest AChE inhibitory activity (58.49 ± 0.95% and 57.33 ± 1.10%), while SOB-hexane extract showed the highest BChE inhibitory activity (46.81 ± 0.75%) and SOB-acetone extract showed the highest tyrosinase inhibitory activity (60.75 ± 0.61%) at a concentration of 200 μg/mL. These results provided new datasets on the major phenolic compounds and biological activities of S. orientalis subsp. bicolor. These results were supported by analyzing the potent antioxidant and enzyme-inhibitory activities of S. orientalis subsp. bicolor.
