Molecular characterization of coriander (Coriandrum sativum L.) genotypes using inter simple sequence repeat (ISSR) and sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) markers

Coriandrum sativum L., belonging to the family Apiceae is an herbal plant and significant for its culinary and medicinal uses. The present study aimed to characterize 30 Coriandrum sativum L. genotypes using protein profiling and inter simple sequence repeat (ISSR) markers. The seeds were collected from Muzaffarabad Division, Pakistan, which consists of three districts: Muzaffarabad, Jhelum Valley, and Neelum Valley. Ten ISSR primers were used in this research, except for one, all primers gave satisfactory results. A total of 30 polymorphic bands and 22 monomorphic bands were observed with an average of 3 polymorphic bands per primer. The largest polymorphism showed by UBC 826 (90%) while UBC857 exhibited the lowest polymorphism (14.2%). The average Polymorphic Information Content (PIC) value observed for all primers was 0.72. A total of 715 amplified bands were produced using the ISSR primers. Clustering of genotypes (CJ) 5 and CJ6 were placed in separate clusters, indicating that they are more genetically diverse. Coriander seeds were used for protein extraction. The Sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) of whole soluble seed proteins exposed a total of 4 bands. A common banding pattern was observed in 22 genotypes with a molecular weight of 15 kilodaltons (kDa), 24 kDa, 36 kDa, and 40 kDa. No band was observed in the 3 genotypes. The results of SDS-PAGE revealed low variation in the coriander genotypes. Additionally, the study’s comprehensive use of ISSR primers to reveal high polymorphism, coupled with the identification of common protein bands, distinguishes it from other research by offering a broader genetic insight into Coriander diversity, particularly focusing on both DNA and protein markers.
