Morphological and molecular characterization of Paratylenchus sacchari n. sp. (Nematoda: Paratylenchinae) from the rhizosphere of Saccharum officinarum in China

Paratylenchus sacchari n. sp. was collected from roots and rhizosphere soil of Saccharum officinarum in Nanning, Guangxi Zhuang Autonomous Region, China. The new species is characterized as follows: female body length 282–383 µm; stylet 16.5–19.2 µm long; lip region with slight constriction or continuous and flat at the frontal end, and with small submedian lobes; excretory pore at the level of the anterior to middle part of the pharyngeal basal bulb; lateral field with three lines; vulval flaps distinct; spermatheca elongate, containing sperm; post-uterine sac absent; tail 15-30 µm long, conical, dorsally convex, and ventrally concave, narrowing suddenly near the terminus forming a non-annulated narrowly rounded tip. Due to similarities in having a stylet less than 40 µm, lateral field with three lines and the presence of advulval flaps, the new species was compared with known species of the genus namely Paratylenchus aquaticus, Paratylenchus leptos, Paratylenchus humilis, Paratylenchus rostrocaudatus, Paratylenchus perminimus, Paratylenchus thysanolus, Paratylenchus vandenbrandei, Paratylenchus chongqingensis. The ITS rDNA and the D2-D3 region of the 28S rDNA gene of the new species were amplified and sequenced. Phylogenetic relationships of this new species with other Paratylenchus spp. were constructed using the ITS rDNA and 28S rDNA D2-D3 gene sequences based on Bayesian inference (BI). A key to the species of Group 2 of the genus Paratylenchus is also provided in this study.
