<p>Grape powdery mildew, caused by <i>Erysiphe necator</i>, poses a significant threat to grape production worldwide. Demethylation inhibitors (DMIs) and quinone outside inhibitor (QoI) fungicides have been widely used for its management. However, reports of resistance to these fungicides have emerged in recent years. This study surveyed major grapevine growing districts of Karnataka state during 2022–2023 to analyze the presence of DMIs and QoIs resistance in <i>E. necator</i>. A modified 24-well leaf-disc bioassay was used to determine the sensitivity of the pathogen to DMIs and QoIs fungicides. The results revealed various levels of resistance among isolates, with mutations in the CYP51 and cytochrome b (cyt b) genes associated with resistance to DMIs and QoIs, respectively. This study reports the first occurrence of DMIs and QoIs resistance in <i>E. necator</i> in Karnataka state, highlighting the need for continued monitoring and alternative management strategies.</p>
