<p>A new xanthone derivative, nodosuxanthone (<b>1</b>), along with six known compounds <b>2–7</b>, were isolated from <i>Calophyllum nodosum</i> to our knowledge for the first time. Their structures were elucidated using various spectroscopic techniques, UV, MS, IR, and 1D and 2D NMR, and confirmed by comparison with the literature data. Compounds <b>1–7</b> were evaluated for their antibacterial activities on five different bacterial strains of <i>Acinetobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Pseudomonas aeruginosa,</i> and <i>Escherichia coli</i> by adapting the well-diffusion method. Among them, the hexane extract, compounds <b>4</b>, <b>5</b>, and <b>7</b>, demonstrated notable antibacterial and bactericidal effects.</p>
