MsGal-2: a key regulator of gill microbiota and its contribution to mucosal immune function in juvenile largemouth bass (Micropterus salmoides)

Galectins, a class of evolutionarily conserved lectins characterized by their affinity for β-galactosides, are multifunctional proteins that mediate critical immune responses, particularly within the realm of pattern recognition and cell–cell interactions. MsGal-2, a prototype galectin, has been identified as a key component in the mucosal immunity of juvenile largemouth bass. This study delves into the immune functions of MsGal-2, focusing on its role in gill microbiota regulation and its opsonic properties. Through high-throughput 16S rRNA gene sequencing, we discovered that MsGal-2 knockdown significantly altered the composition of the gill microbiota, underscoring its crucial role in maintaining microbial diversity. In vitro experiments revealed that recombinant MsGal-2 (rMsGal-2) selectively bound to certain microbes, particularly those isolated from largemouth bass, and enhanced leukocyte encapsulation, indicating its opsonic nature. These findings not only highlight the multifaceted immune functions of MsGal-2 but also illuminate the intricate interplay between host immunity and the microbiome in aquatic species.
