Functional activities of Lacticaseibacillus casei in culture of Oplegnathus punctatus

This study investigated the survival dynamics of Lacticaseibacillus casei YB-38 in nutrient-rich seawater and its inhibitory effect on the pathogenic bacterium Vibrio harveyi YL-10. L. casei YB-38 at 108 CFU per gram of feed was supplemented with feed in the cultivation of Oplegnathus punctatus (initial average weight = 5.30 ± 0.20 g), focusing on the growth characteristics for 14 days. Flow cytometry and genomics analysis were utilized to investigate the effects of L. casei YB-38 on the intestinal microbiota activity and immune function of O. punctatus. The results indicated that L. casei YB-38 and V. harveyi YL-10 demonstrate enhanced growth under simulated enriched conditions. V. harveyi YL-10 exhibited better growth when attaching feed particles in seawater, irrespective of feed addition. L. casei YB-38 at a concentration of 10⁷ CFU/mL inhibited V. harveyi YL-10, which was initially present at 103 CFU/mL, and the inhibition lasted for 8 h. The addition of L. casei to the feed significantly enhanced the growth of O. punctatus, and modulated the gut microbiota by reducing the relative abundance of the genus Vibrio and increasing that of Lactobacillus. O. punctatus fed with L. casei YB-38 exhibited smaller fluctuations in intestinal microbiota compared to the control group, indicating the stabilizing effect of L. casei YB-38 on the intestinal microbiota. Furthermore, the inclusion of L. casei YB-38 in the feed potentially bolstered the growth metabolism of O. punctatus, including pathways related to protein digestion and absorption, and antigen processing.
