Evaluation of dietary fermented Gracilaria on growth, whole-body composition, intestinal insulin-like growth factor 1 (IGF-1) and α-amylase mRNA expression, and intestinal histology of golden rabbitfish Siganus guttatus

Gracilaria is a promising locally feed ingredient for golden rabbitfish, Siganus guttatus in Indonesia. A study was conducted to evaluate the effect of fermented Gracilaria (FG) inclusion at 0% (CTRL), 20% (FG20), 25% (FG25), and 30% (FG30) in formulated diets on growth, whole-body composition, intestinal IGF-1 and α-amylase mRNA expression, and intestinal histology of golden rabbitfish. All test diets were isonitrogenous with 34% crude protein and 9–11% crude lipid. Juvenile golden rabbitfish were stocked in fiber tanks and fed for 60 days. Fish fed the FG20 and FG25 diets exhibited significantly higher SGR and lower FCR than the control diet. All FG diets significantly increased whole-body protein and amino acid content compared to the control. The intestinal mRNA expression of IGF-1 showed a significant increase of 2.2-fold change in fish fed the FG25 diet and 1.2-fold change in fish fed the FG20 diet. In the anterior intestine, fish fed the FG25 diet showed moderate alterations including necrotic on mucosa, villi, and submucosa while fish fed the FG30 diet displayed significant alterations with necrotic villi, submucosa, and muscularis. In the posterior intestine, FG25 had minimal alterations with villi hyperplasia, while fish fed the FG30 diet exhibited considerable alterations with hyperplasia of goblet cells and necrosis of villi. Based on the growth performance, whole-body composition, intestinal IGF-1 mRNA expression, and intestinal histology results, we conclude that FG can be included in diets for golden rabbitfish at up to 20% without negatively impacting intestinal structure. The study supports the development of local aquafeed and enhances the value chain for Indonesian seaweed farmers.
