Comparative reproductive performance and gonadal transcriptomics of mature males and neo-males in yellow drum (Nibea albiflora)

Yellow drum (Nibea albiflora) is a commercially important marine fish distributed widely in the coastal waters of China, Japan, and Korea. Our previous research has successfully established technology for inducing sex reversal neo-males, yet the detailed characterization of sperm quality and molecular pathways in males and neo-males remains unclear. Here, we investigated the reproductive characteristics between males and neo-males through analysis of serum sex hormone levels, sperm quality, and comparative transcriptomics of gonads. The analysis of serum sex hormone levels showed no significant differences between males and neo-males, except for FSH in the late-spermiation stage and GTH in the post-spermiation stage. Moreover, sperm quality also exhibited no significant difference. These findings suggest comparable reproductive performance in sex-reversed neo-males. Furthermore, comparative transcriptomics identified a total of 5792 differentially expressed genes (DEGs) in the mature male vs neo-male group, including 4536 upregulated and 1256 downregulated genes. GO (gene ontology) analyses revealed signaling in biological processes, cell surface in cellular components, and receptor binding in molecular function were involved in regulating male testis development. KEGG pathway analyses revealed that cytokine-cytokine receptor interaction may play important roles in the development of neo-male testes. Overall, our findings indicate that although hormone levels and sperm quality are largely similar, subtle differences in reproductive regulation exist between males and neo-males. This study provides key insights into the physiological and molecular characteristics of neo-male yellow drum, enhancing our understanding of their reproductive biology.
