<p>Endophytic fungi are mutualistic microorganisms that live within plant tissues without harming the host. They enhance plant health and are valuable sources of bioactive secondary metabolites for medicinal use. This study aimed to identify the endophytic fungi in the medicinal plant <i>Catharanthus roseus</i>, collected from different locations in Jharkhand, India. A total of thirty unique fungal isolates were obtained and maintained through continuous subculturing. Among them, <i>Muyocopron dipterocarpi</i> has been selected for further chemical profiling based on its promising bioactive potential. Primary screening of the metabolite was done through alkaloid test and HPTLC analysis. A comprehensive metabolite profiling of the selected isolate has been performed by employing advanced analytical techniques such as high-performance liquid chromatography, gas chromatography-tandem mass spectrometry, and liquid chromatography-high resolution mass spectrometry. The GC–MS/MS analyses of extracts from each species identified multiple bioactive compounds, including 1-tetradecene, 2,4-Di-tert-butylphenol, 1-hexadecanol, ethyl iso-allocholate, digitoxin, and 1-hexacosene. Using LC-HRMS for comprehensive profiling of fungal metabolites, we identified several indole alkaloids from <i>Muyocopron dipterocarpi</i> for the first time, including gelsemicine, ajmaline, brassitin, and geissospermine. Among the metabolites detected, ajmalicine was found to be the predominant monoterpenoid indole alkaloid present in the crude extract. Structural confirmation was established via Orbitrap high-resolution mass spectrometry, which provided evidence of a unique sodium adduct ion at m/z 375.1686, which matches the theoretical mass. Additionally, HPLC quantification of ajmalicine indicates that it is the primary indole alkaloid in the selected groups, with an estimated concentration of 106.1&#xa0;µg/mL.</p>
