Eutypella platanica sp. nov. and Other Fungal Genera Cause Oriental Sycamore (Platanus orientalis) Tree Decline in Urban Forests of Iran

Urban forest trees provide numerous benefits, including improving air quality, reducing urban heat, and enhancing biodiversity. They offer shade, which lowers energy costs and contributes to mental well-being by creating green spaces. Canker, dieback, and decline are major fungal diseases impacting woody plants, leading to substantial economic losses in urban forests. This study aimed to classify and evaluate the pathogenicity of fungus species linked to these issues in oriental sycamore trees in Tehran Province, Iran. A total of 20 Diatrypaceae isolates were obtained from oriental sycamore trees displaying symptoms of stem canker, decline, branch dieback, and defoliation. Additionally, molecular identification using the ITS region revealed various fungi from Ascomycota and Basidiomycota associated with tree decline. Morphological assessments and phylogenetic analysis of the ITS and tub2 regions identified a new Diatrypaceae species, Eutypella platanica sp. nov. Pathogenicity tests revealed that E. platanica was responsible for symptoms of canker, dieback, and decline. Additionally, by conducting pathogenicity tests, E. platanica isolates showed that higher temperatures (28 °C) led to significantly longer average lesion lengths, indicating a more pronounced pathogenic impact than lower temperatures (24 °C). This study identified E. platanica as a newly discovered species and the causal agent of canker and dieback in Platanus orientalis, presenting a significant emerging threat to the health and sustainability of urban forests in Iran. Taken together, our results provide the foundation and basis for future epidemiological research and the development of management strategies.
