Bacterial Communities Associated with Litchi Leaf Roller, Statherotis leucaspis (Meyrick, 1902) (Lepidoptera: Tortricidae) Reared On Different Litchi Varieties

The leaf roller, Statherotis leucaspis Meyrick, has emerged as the dominant pest species, inflicting considerable yield losses on litchi in India. The chemical composition of different host plant varieties can modulate symbiotic bacteria in the gut, leading to growth and development. The present study examines the biochemical compositions of litchi varieties, i.e., ‘Shahi’, ‘Swarna Roopa’, and ‘China’, and the comparative structure and diversity of gut microbiota of S. leucaspis larvae reared on different varieties using 16S rRNA amplicon sequencing. The ‘Swarna Roopa’ variety of litchi showed the highest resistance against S. leucaspis, followed by ‘Shahi’, and the highest susceptibility in ‘China’ in terms of infestation, growth, and development. The results revealed that S. leucaspis infestation was strongly correlated (p < 0.05) with phenol, flavonoid, free amino acid, and carbohydrate contents in litchi leaves. Gut microbial analyses revealed that Proteobacteria and Firmicutes were the dominant phyla with high relative abundances in ‘Shahi’ and ‘China’. The relative abundance of Actinobacteria, Acidobacteria, and Planctomycetota was noted to be higher in highly resistant (‘Swarna Roopa’) and resistant (‘Shahi’) litchi varieties. Principal component analysis (PCA) demonstrated a distinct group of tested varieties with associated bacterial phyla and biochemical compositions. Overall, the findings indicated that the biochemical variability of litchi varieties significantly altered the gut bacterial diversity of S. leucaspis, providing insights into the development of novel pest management strategies.
