Rapid, simplified LAMP assay to detect Agroathelia rolfsii (syn. Sclerotium rolfsii) in black soybean with damping off

A loop-mediated isothermal amplification (LAMP) assay that was developed for Agroathelia rolfsii (syn. Sclerotium rolfsii) gave a positive reaction for A. rolfsii, but not Calonectria ilicicola or Phytophthora species. The LAMP reaction with 100 fg or more of A. rolfsii DNA was completed in 1 h. The LAMP assay was 1000 times more sensitive than conventional PCR and detected A. rolfsii-infected soybean seedlings in fields. The technique provides a rapid, simplified diagnosis for disease management.
