Lysine inhibition of expression of genes encoding type III secretion system in Ralstonia pseudosolanacearum and its virulence in host plants

The type III secretion system (T3SS) is essential for Ralstonia pseudosolanacearum to infect host plants. Transcription of the T3SS genes was not activated in nutrient-rich casamino acid-peptone-glucose (CPG) broth, but activated in a minimal medium that might mimic metabolic signals of the host plant. CPG broth at 5% substantially impaired expression of T3SS genes. In tests of T3SS gene expression, l-lysine was found to be a novel inhibitor of expression of T3SS genes in non-acidic conditions and significantly impaired their expression via a PrhG-HrpB pathway and their motility and biofilm formation, which are also important for pathogenicity, and led to significant inhibition of symptom development in tomato plants.
