Lamotrigin plus Valproat: Mythos oder rationale Kombinationstherapie?

A significant number of people with epilepsy are treated with a combination therapy of antiseizure medications. This raises the need to identify appropriate drug combinations. The combination of lamotrigine and valproate is particularly commonly recommended. This review examines the extent to which these recommendations reflect a myth, i.e., a traditional recommendation with insufficient data, or a well-founded, rational combination therapy. On the basis of a large number of case histories, retrospective and prospective studies, supported by preclinical data, it is shown that the combination of lamotrigine and valproic acid can be considered a rational combination therapy. Practical recommendations for the use of this drug combination are given, and open questions and possible perspectives are pointed out.
