<p>An important first step in isolating the bioactive ingredients from medicinal plants is extract preparation. Numerous parameters influence the separation of biomolecules, and even slight changes in their values may have an impact on the extraction process. Therefore, it becomes essential to accurately check the impact of every parameter. Quality by design is becoming a popular method for creating a process that results high-quality output. Following single-factor research and response surface technique, the current work develop, optimize, and validate an extraction process for lupeol extraction from <i>Uraria picta</i> whole plant using analytical quality by design. Here, the content of lupeol was examined using a validated thin-layer chromatography (TLC)‒densitometry approach. According to the investigations, solvent volume and ethanol percentage have separate effects on the extraction process. These were chosen, and Central Composite Rotatable Design (CCRD) further optimize their impact. The model’s fitness was evaluated using ANOVA (analysis of variance) based on regression coefficients and additional quality evaluation metrics. For the maximal extraction of lupeol by reflux technique, the produced information suggests using 50&#xa0;mL/g of 80% ethanol for 30&#xa0;min (× 2 cycles of reflux) with a desirability of 0.927. All extraction procedures for plant standardization and the production of herbal formulations of <i>U</i>. <i>picta</i> may benefit from the designed extraction protocol. A discriminating analysis between the various parts of <i>U</i>. <i>picta</i> was also included in this study, and the results showed that the stem and leaves have a higher concentration of lupeol than other parts. Hence, can be used in place of whole plant to conserve this plant.</p>
