Sawtooth thin-layer chromatography coupled with electrospray ionization mass spectrometry for detection of adulterants in traditional herbal medicines

Coupling thin-layer chromatography (TLC) with mass spectrometry (MS) is a useful technique for the detection of active constituents and adulterants in functional foods, drinks, and herbal products. In this study, sawtooth TLC plates were prepared for TLC‒MS detection of adulterants in herbal medicines. A high-voltage power supply (+ 5 kV) was connected to induce electrospray ionization (ESI) without the need to detach the TLC gel bed. Each analyte separated on the TLC plate was dissolved at the triangular tips and ionized by ESI. To test the analytical efficiency of the method, six most commonly used adulterated compounds were added to commercially available herbal powder samples followed by compound extraction and separation on a silica TLC plate. After drying, the TLC plate was cut into two sawtooth plates and then installed on the XYZ stage, and the analytes at the triangular tips were sequentially scanned and detected with ESI–MS. The results indicated that the sawtooth TLC‒ESI–MS method successfully detected the adulterants in the chemically complex herbal samples.
