Preparation and characterisation of Lidocaine elastic microneedles for local anaesthesia

The burning or stabbing pain caused by postherpetic neuralgia (PHN) complications severely impacts patients’ quality of life. Current treatments often involve analgesics or antiviral medications, with anaesthetic intervention targeting nerve conduction being a common approach for pain relief. Common anaesthetic methods include general anaesthesia, regional anaesthesia, and local anaesthesia. While local anaesthesia is widely used for postherpetic neuralgia, clinical challenges persist, including low efficacy and high patient discomfort. This study proposes a sustained-release lidocaine elastic microneedle for local anaesthesia, featuring a specially designed bullet-shaped microneedle and array structure. Following systematic characterisation of its morphology, dimensions, and mechanical properties, its swelling behaviour, mechanical performance, and transdermal drug delivery capabilities were further investigated. Experimental data indicate that the CS/PVA/LD/DBA-MN bullet-shaped microneedles exhibit excellent forming effects and mechanical properties capable of penetrating the skin stratum corneum. The microneedles released 59.66% of the loaded drug within 2 min, with complete drug release achieved within 72 h and a drug diffusion efficiency of 93.23%. Compared to traditional conical microneedles, they exhibited lower penetration resistance, superior swelling effects, and higher drug release efficiency, offering a novel technical approach for local anaesthetic delivery.
