<p>Type one protein phosphatases (PP1) participate in multiple biological processes such as cell cycle regulation, development, signaling of hormonal pathways, and stress response in eukaryotes. Pleiotropic functions of PP1s are modulated through their interaction with positive or negative regulators which control either their subcellular localization or the substrate specificity to orchestrate diverse signaling pathways. PP1s have been intensively studied in mammals due to their involvement in many diseases but the functions of PP1s in plants remain largely unexplored. In this review, we provide a landscape of the molecular and structural properties of PP1s in the plant model <i>Arabidopsis thaliana</i> and in crops, highlighting their role in the regulation of hormonal and stress signaling pathways. Moreover, we provide the comprehensive interactome of plant PP1s and complete the identification of <i>PP1</i> genes in cereals, ranging from bread wheat to durum wheat and barley.</p>
