<p>Ran (Ras-related nuclear) proteins are small monomeric GTPases and regulate vital cellular processes viz. nucleocytoplasmic transport, mitotic spindle assembly, nuclear envelope re-assembly, and cell cycle regulation. The study describes functional characterization of a <i>RAN1A</i> promoter from wheat by promoter deletion (-1970, -1446, -415&#xa0;bp) analysis in tobacco. The spatio-temporal regulation during development, and response to hormones and abiotic stresses was analyzed by GUS histochemical and quantitative expression analyses. The GUS staining revealed that <i>TaRAN1A</i> was ubiquitously expressed in vegetative tissues. However, at reproductive stage, <i>TaRAN1A</i> exhibited an organ- and tissue-specific expression which was developmentally regulated. The GUS staining was localized at the base of the calyx, while there was no expression in petals. In stamens, GUS staining was observed in mature anthers at fixation point while in pistil the juvenile stigma exhibited significantly higher expression. Similarly, significantly higher level of <i>TaRAN1A</i> expression was recorded in mature seeds. There was no visible difference in the spatio-temporal profile of three promoter fragments during development. Except that the full-length (1970&#xa0;bp) region of <i>TaRAN1A</i> promoter exhibited higher expression as compared to its deletion fragments (-1446 and -415&#xa0;bp). The roles of different <i>cis</i>-regulatory regions were more evident under hormone and abiotic stress treatments. The three promoter fragments showed differential root- and shoot-specific expression and inducibility in response to ABA and cold. In conclusion, <i>TaRAN1A</i> exhibits a tissue-specific regulation during development, hormone treatments, and abiotic stress conditions. Our study indicates a potential role of <i>TaRAN1A</i> in reproductive organ development.</p>
