Allyl isothiocyanate as a natural fumigant for protecting soybean and bean grains against Aspergillus parasiticus growth and aflatoxin production

This study evaluated the efficacy of gaseous allyl isothiocyanate (AITC) in inhibiting Aspergillus parasiticus growth and aflatoxin production in soybeans and beans stored at 85% relative humidity for 35d. Samples of 50 g of soybeans or beans were inoculated with A. parasiticus (10⁴ spores/g) and treated with 0, 1, 5, or 25 µL/L of gaseous AITC in hermetically sealed glass jars. After 35d, microbial counts were determined on potato dextrose agar, and aflatoxins were extracted, concentrated using immunoaffinity columns, and quantified via high-performance liquid chromatography with fluorescence detection. The moisture content of the grains was also monitored throughout the storage period. The results showed that AITC at 5 and 25 µL/L reduced A. parasiticus by 2 and 4 Log CFU/g in soybeans, respectively. In beans, reductions of 2 and 5 Log CFU/g were observed at the same concentrations. Aflatoxin B1 (AFB1) production was completely inhibited at 25 µL/L and reduced by approximately 90% at 5 µL/L. Gaseous AITC at 1 µL/L was ineffective in inhibiting fungal growth in soybeans and did not reduce AFB1 production in either crop. During storage, the moisture in soybeans increased from 14 to 18%, while the moisture in beans rose from 14.5 to 19%. These findings suggest that AITC fumigation may be a viable strategy to prevent A. parasiticus growth and AFB1 contamination in stored soybeans and beans, potentially improving food safety and grain preservation.
