<p>Quorum-sensing agonists significantly enhance biofilm formation and bioleaching efficiency. In this study, we synthesized acyl-homoserine lactone (AHL) analogs and investigated their effects on biofilm formation in <i>Acidithiobacillus thiooxidans</i>, mineral adsorption kinetics, and pentlandite bioleaching. The results showed that A-3 promoted biofilm formation, enhanced adsorption capacity toward pentlandite, and improved pentlandite bioleaching efficiency. Molecular docking revealed that A-3 exhibited higher binding affinity for the quorum-sensing receptor protein AfeR than its natural signaling ligand. In summary, compared to the natural signaling molecule, A-3 significantly enhanced biofilm formation and adsorption capacity, consequently improving bioleaching efficiency. This establishes a theoretical foundation for optimizing bioleaching in industrial applications.</p> Graphical abstract <p></p>
