Fine mapping of the Co-Bf gene conferring broad-spectrum resistance to the anthracnose pathogen of common bean (Phaseolus vulgaris L.)

AbstractGenetic resistance is considered the most effective strategy to manage Colletotrichum lindemuthianum, a pathogen with many virulent strains that cause the anthracnose disease of the common bean (Phaseolus vulgaris L.). In this study, we inoculated the Andean common bean cultivar Beija Flor (Bf) with six Andean and nine Mesoamerican races of C. lindemuthianum. Beija Flor was resistant to 89% of the Mesoamerican and to 50% of the Andean races of the pathogen. The inheritance of resistance study of Beija Flor, using F2:3 families and F2 plants from three populations derived from two different crosses, revealed the presence of a single dominant resistance gene in Beija Flor. We then performed the fine mapping of this gene. F2 plants from two different crosses were used to perform bulked segregant analysis with the BARCBEAN12K_BeadChip, which includes 11,292 single-nucleotide polymorphisms. Fine mapping was achieved using F3 plants derived from the Beija Flor × Crioulo 159 cross and genotyped with Kompetitive Allele-Specific PCR (KASP) markers. The resistance gene in Beija Flor, previously designated Co-Bf, was fine mapped to a 30.2 Kb genomic region on chromosome Pv04, between KASP markers SS333 (342,926 bp) and SS509 (373,114 bp). Three genes were annotated within this genomic region, including the Phvul.004G005600.1, a gene associated with plant disease resistance. The KASP markers identified in this study could be utilized in marker-assisted selection to pyramid the Co-Bf with other resistance genes into a single cultivar, thereby enhancing durable resistance in common bean against C. lindemuthianum.
